What is it about?

A brief review on direct methods in protein crystallography is given. Examples are also given on difficult SAD phasing, model building and low-resolution phase extension.

Why is it important?

A direct method has been incorporated in an iterative dual-space phasing and model building framework. The direct method is acting as a phase restrain procedure in the reciprocal space. This makes the iteration particularly powerful in difficult SAD phasing, model completion and low-resolution phase extension.
